Explain sigmoidal nature of oxygen dissociation curve for hemoglobin.

Hemoglobin’s oxygen dissociation curve is sigmoidal, reflecting its cooperative oxygen-binding ability and efficiency in transport. Hemoglobin is a tetramer, with four subunits, each capable of binding one oxygen molecule. The curve plots oxygen saturation versus partial pressure of oxygen (PO2). At low PO2, such as in tissues, hemoglobin binds oxygen weakly, allowing easy release. As PO2 rises in the lungs (~100 mmHg), binding of one oxygen molecule induces conformational changes that increase the affinity of the remaining subunits, producing the characteristic S-shaped curve. This cooperativity ensures efficient oxygen uptake in the lungs and release in metabolically active tissues (~40 mmHg). Factors such as low pH, elevated CO2, increased temperature, or high 2,3-bisphosphoglycerate (2,3-BPG) shift the curve to the right, promoting oxygen unloading where it is most needed, like in exercising muscles. This finely tuned mechanism allows hemoglobin to balance oxygen loading and delivery, adapting to varying tissue demands and maintaining effective oxygen supply throughout the body.
