The Golgi apparatus functions like a cellular post office, meticulously sorting and directing proteins to their appropriate destinations. Proteins synthesized in the rough endoplasmic reticulum (ER) are transported to the Golgi in small vesicles. Structurally, the Golgi is organized into a series of flattened membrane-bound sacs called cisternae—cis, medial, and trans—through which proteins pass sequentially. During this transit, proteins undergo critical modifications, such as glycosylation, where sugar molecules are added, or phosphorylation, which can influence protein activity or localization. Each protein is tagged with a molecular “address label” to indicate its final destination, such as mannose-6-phosphate for lysosomes or signal sequences for secretion. Vesicles budding from the trans-Golgi network carry the modified proteins to lysosomes, the plasma membrane, or the extracellular space. Receptors recognize these labels, ensuring precise delivery. Any disruption in this sorting process can lead to mislocalization of proteins and cellular dysfunction, highlighting the Golgi’s essential role in maintaining cellular organization and proper physiological function.
