1. Activation:

  • Occurs in lymph nodes when antigen-presenting cells (APCs), such as dendritic cells, display pathogen-derived peptides on MHC class I molecules.
     
  • The T-cell receptor (TCR) recognizes these antigen-MHC I complexes.
     
  • Co-stimulatory signals like CD28 binding to B7 molecules on APCs are essential for full activation, ensuring the T-cell responds only when necessary.

2. Target recognition:

  • Activated cytotoxic T-cells circulate and scan body cells for the same antigen-MHC I combination.
     
  • Tight binding ensures specificity, reducing damage to healthy cells.
     

3. Killing mechanism:

  • Perforin forms pores in the target cell membrane.
     
  • Granzymes enter through these pores, triggering apoptosis by activating caspases, which dismantle cellular structures and DNA.
     

4. Cytokine secretion:

  • They release interferon-gamma (IFN-γ), enhancing immune responses and recruiting other immune cells.
     

5. Role in health:

  • They eliminate virus-infected cells, preventing viral replication.
     
  • They recognize and kill tumor or transformed cells, contributing to cancer surveillance.
     
  • This highly specific killing mechanism ensures the immune system destroys compromised cells while sparing healthy tissue.
     

Through this precise and regulated process, cytotoxic T-cells maintain immune balance, prevent infections from spreading, and safeguard the organism from malignancy.
