Steps in Plant Protoplast Culture:
Plant protoplast culture involves isolating and culturing plant cells without their cell walls.

  1. Tissue Selection: Healthy, metabolically active tissues (e.g., young leaves, calli) are chosen.
  2. Sterilization: Tissues are surface-sterilized to prevent contamination.
  3. Enzyme Treatment: Tissues are incubated in an osmoticum containing cell-wall-degrading enzymes (cellulase, pectinase, hemicellulase) to remove the cell wall, releasing individual protoplasts.
  4. Protoplast Isolation & Purification: Protoplasts are gently separated from cellular debris, typically by centrifugation and washing, and collected in an osmotically adjusted medium.
  5. Culture: Purified protoplasts are cultured in a liquid or semi-solid nutrient medium, which maintains osmotic pressure and provides essential nutrients for survival and cell wall regeneration.
  6. Regeneration: The protoplasts regenerate a cell wall, divide to form callus, and eventually differentiate into complete plants under specific hormonal regimes.

Major Limitations:

  1. Low Regeneration Efficiency: Many plant species or genotypes are recalcitrant, failing to regenerate into whole plants from protoplasts.
  2. Somaclonal Variation: Genetic changes can occur during culture, leading to undesirable variations in regenerated plants.

Role of Somatic Hybridization in Crop Improvement:
Somatic hybridization involves fusing protoplasts from two different plant species or genera to create somatic hybrids. This technique is crucial for crop improvement by:

  • Overcoming Sexual Incompatibility: It allows genetic exchange between sexually incompatible species, introducing desirable traits (e.g., disease resistance, stress tolerance) from wild relatives into cultivated crops.
  • Creating Novel Hybrids: It generates unique genetic combinations not possible through conventional breeding, potentially leading to new varieties with improved traits.
